In Los Angeles, police estimated 100,000 people had joined a march led by singer Gloria Estefan.
"It's the right of every American to protect where they live," she told the crowd.
"But that doesn't give them a reason to place a law that could create racism and discrimination."
The BBC's Rajesh Mirchandani, at the rally, says there were banners calling for a boycott of Arizona, and even one portraying the state's governor, Jan Brewer, as Hitler.

Hey, BBC, you liberal jerks! Earlier reports said the police estimated the crowd to be maybe 25,000.
I spent a couple of nights at the El Tovar on the South Rim of the Grand Canyon back in the early 1990s. Fantastic place and great dining service and food. A place where Teddy Roosevelt stayed. Huge wooden beams across the ceilings. ...No elevators or central air, but basically what today is considered antique furniture in the tiny bedrooms.
And if someone comes into our home uninvited, we get to throw them out. Why?
Because there IS NO RIGHT to tresspass.
