Those destroyers could be anywhere in the world; only thing that would be sweeter would be able to intercept from a sub. Not that I think this is beyond the abilities at the moment, but having a sub launch the intercept would send chills down anyone’s back - you could block any medium range missile anywhere in the world, and no one would know where your platforms were.

How much prep time does a sub need to fire a missile? It’s certainly doable.
