He really could not have given a more perfect response.
That is, if Speaker Ryan wins his primary, wins reelection, the Republicans win the House again, and Donald Trump wins the Presidency. The next step is Ryan's primary, and Trump carried his district in the Presidential primary so this was an unforced error on Ryan's part.
Trump is doing exactly the right thing.
As we watch the GOP elite suffer apoplexy from the anticipation of losing their gravy train for good, the People are taking charge.

PAUL RYAN is such a POMPOUS PRICK. He needs to put his sense of self-importance where the sun doesn’t shine.
Instead Ryan adds TWO TRILLION to the budget and the deficit.
And Ryan has the unmitigated gall to say, and I paraphrase, "We've got to make sure we have a candidate who believes the conservative principle of LIMITED GOVERNMENT."
And THAT'S just ONE example. It's all ass backwards. RYAN was espousing WORDS that vaguely SOUND conservative, he and Repubs in DC do NOTHING to really make those words HAPPEN, then Trump's entire PLATFORM is based on the full honest-to-gosh things that Ryan criticizes Trump for!
Another example. Ryan says we've got to make sure the Party upholds the constitution. WHO HAS BEEN THE STRONGEST on the SECOND AMENDMENT? Trump. IN EVERY SPEECH. "The 2nd Amendment will be STRONGER THAN EVER in a Trump administration."
Trump questioned if Obama was constitutionally qualified to be president. Trump question the same thing with Cruz. So WHO defends the constitution????
