However, we have not been able to isolate what Covid 19 looks like. The PCR tests only look for symptoms.
So how do they know there is a “variant” from a virus they have not been able to identify?

Always fighting the last war. The Delta Variant is here and spread fast already. The Colombian variant is the next big thing. 10% of all new cases. Given the name, it isn’t coming here from the EU.
But illegal aliens swarming over the southern border are just fine.
