For the first time, a researcher has determined that the naturally occurring dietary supplement known as nicotinamide riboside (NR) can enter the brain.
The discovery was made by Christopher Martens and Dr. Dimitrios Kapogiannis. The finding is significant because it supports the idea that NR, upon reaching the brain, can alter the metabolism of relevant biological pathways involved in neurodegenerative diseases like Alzheimer's.
Upon consumption, NR is readily converted into nicotinamide adenine dinucleotide (NAD+), which is critical to cellular repair and the repair of damaged DNA.
"NAD+ is gradually lost as we get older or develop chronic diseases. Loss of NAD+ is linked to obesity and other negative lifestyle habits like smoking," Martens said.
"We had some preliminary signs of efficacy, including lower blood pressure in people who had high blood pressure to begin with," he said.
Martens and colleagues measured NAD+ directly in tiny particles called extracellular vesicles that originated from neurons and ended up in the blood.
The researchers determined, first, that NAD+ levels went up in these vesicles after six weeks.
"When NAD+ goes up in these vesicles, we see an association with some of the biomarkers of neurodegenerative disease," Martens said. "Particularly, in people where we saw an increase in NAD+, we also saw changes in biomarkers like amyloid beta and tau, which are both related to Alzheimer's disease," Martens said.
Nearly all drugs for patients with Alzheimer's have only a modest effect on the symptoms but do not significantly stop the underlying progression of the disease.
"In our ongoing trial, we're measuring markers of cognitive function and other things related to functional independence and quality of life, but we're also hoping to gain some insight on the underlying disease process," Martens said. "We're hoping that the people who take the NR might have preserved function."

NMN should work the same. I think that is also easier to find.

I quit smoking some 25 years ago. But maybe the nicotine of the nicotinamide of the Skoal I still chew will have a redeeming quality.
I know, I know it maybe unhealthy. Just saying. It’s a habit, but as Twain said “quitting tobacco is easy, I’ve done it many times.”

I have been researching the use of NR and the other NAD+ precursor, NMN, for a few months now. The only definitive studies that showed tissue absorption levels have been done on mice. Both NR and NMN were tagged with a radioactive marker so that they could be detected in the mouse tissue when absorbed.
The studies showed that NR and NMN that was orally ingested did not show significant uptake levels in body tissues like the liver, kidneys, or brain. Injecting the mice with the supplements did show up in their tissues, but only at dosage levels that were well above what would be considered safe for humans.
Amazon has recently banned the sale of NMN due to a large amount of fake products being marketed. If you want to biohack these the safest companies would be Tru Niagen and DoNotAge. DoNotAge is made in the UK and is a fraction of the cost of Tru Niagen.

Actually, nicotine and nicotinamide are very similar chemically. They both bind to specific cell receptors that enable transferring the molecules into the cell. The problem is that when nicotine, which is harmful to cell DNA, binds to cell receptors it blocks nicotinamide (NR, NMN), which is beneficial to the body and slows ageing, from entering the cell. This is one of the reasons people who smoke often appear to age prematurely. They are.

LOL! I know what you mean. We used to make fun of Dad at the table with his bottles of supplements standing at attention like little soldiers around his plate. I am not laughing any more.
Yeah - does not surprise me. It still seems that one cannot rationally argue smoking tobacco has any benefit whatsoever to health, but only a legion of risks.

Many do the same thing.
Pregnenolone makes beta carotene last longer. Beta carotene makes vitamin c last longer. So these 3 can have a similar effect.
Potassium makes calcium last longer.
Sodium makes magnesium last longer. Magnesium makes some vitamins last longer.
The body can use tryptohan, a protein part, to make niacin, so consuming foods with protein can have a similar effect to taking niacin.
People mix retinol with skim milk. Grocery stores sell that milk. Many people have excess retinol. Smoking, drinking beer, yogurt, being in the sun reduce excess retinol.
If people want to consume something it’s because it has a benefit.

The NAD+ that I take is liposomal and includes a betaine for better absorption. No, I don’t have rigorous bio-assay proof of that.

There appear to be “hundreds” of claimed NMN products for sale on Amazon right now.
