Having that second cup may actually be good for coffee drinkers, according to a discussion of coffee's preventive and therapeutic benefits to human health in a recent review.
Research showed the mechanisms of action of coffee are complex and dependent on the effects of its constituents, including chlorogenic acids, polyphenolics, terpenoids, alkaloids and other phytochemicals.
"We also found evidence that the antioxidant activity of coffee, which activates the nuclear factor erythroid 2-related factor 2, or Nrf2, may be an important mechanism of action," Davidson said.
They found evidence protective effects of coffee in the gut decreased colon cancer risk, which may be due to its activity as an aryl hydrocarbon ligand. The aryl hydrocarbon receptor, AhR, is a transcription factor that regulates gene expression.
The study also demonstrated that roasted coffee-derived extracts function in part by activating the AhR. Coffee induced several AhR-dependent responses in the intestine. These included gene expression, inhibition of intestinal stem cell-enriched organoid growth and inhibition of intestinal barrier damage.
Their research also found that some coffee components bind the orphan nuclear receptor NR4A1 to the interactions with the AhR receptor. The NH4A1 receptor is a key factor in multiple diseases, such as arthritis, inflammation, cancer and cardiovascular diseases, and a high NR4A1 expression is associated with breast cancer.
The paper also refers to research focused on developing and discovering compounds for mechanism-based drug development to target both cancer and non-cancer endpoints.
"A major target for cancer chemotherapy includes specific protein transcription factors, the aryl hydrocarbon receptor, the estrogen receptor and orphan nuclear receptors NR4A1," Safe said.
Research has included collaboration to investigate the role of the aryl hydrocarbon ligand and its microbial metabolites on intestinal function and disease.
"We also have ongoing collaborations focused on endometriosis, Parkinson's Disease, and learning and memory," Safe said.

Given what we know about who pays for various studies, should I ask if big coffee paid for this?

But here’s the way it usually goes:
Study #1: Coffee is good for you.
Study #2: No. Coffee is bad for you.
Study #3: No. Coffee is good for you.
Study #4: No. Coffee is bad for you.
Etc.
Sometimes I wonder if these guys question the current wisdom just to get published.

I drink two cups of strong coffee with heavy whipping cream daily....I am a firm believer it is GOOD for me!!...

My reaction is a little different.
My first thought is of the experts who a few years ago insisted that it would kill you.
I think they were the same ones that told us that they knew all about the dangers of cholesterol.
I wonder what mavels they will have for us next?
I typically drink coffee “black as midnight on a moonless night,” to quote David Lynch (I think).
But occasionally I mix a cup into a vanilla milkshake. Damn, that tastes good.
I roast in a good stove top popcorn popper. What’s your roasting technique?
Now that I no longer work, I’m drinking 4-5 cups daily. In a large mug. But, I’m buying the cheap stuff since the crappy brands now cost so much! Hope I’m healthy.
