Researchers have unveiled crucial insights into the factors that might influence Intervertebral Disk Degeneration (IDD).

IDD is a predominant cause of lower back pain, impacting millions worldwide. The focus of this research revolved around nucleus pulposus cells (NPCs), pivotal in IDD, and how oxygen levels and the HIF1A gene could influence them. Employing advanced RNA sequencing (RNA-seq), the team meticulously captured a comprehensive transcriptome of NPCs exposed to different oxygen conditions and instances of HIF1A deletion.

The study leveraged tissues sourced from patients who had given informed consent before undergoing surgery. The efficacy of HIF1A modifications was authenticated using q-PCR, leading to a deeper understanding of the gene expressions under varying conditions. Significantly, the team spotlighted the potential role of oxygen concentration and the HIF1A gene in determining the behavior of NPCs, particularly emphasizing its implications on lipid metabolism.

These insights might provide a more comprehensive understanding and inform novel therapeutic approaches for IDD in the future. However, as is the norm in scientific discovery, while these results shed light on mechanisms of IDD and NPCs, further investigations are essential to validate the findings and translate them into clinical applications.

It appears oxidized LDL is quite instrumental in the destruction of our back discs. Minimizing LDL and/or oxidized LDL, could be helpful.

From the published paper:

“The presence of ox-LDL and LOX-1 was positively related with the extent of IVDD in nucleus pulposus (NP), end-plate cartilage and outer annulus fibrous, but not with the extent of degeneration of inter annulus fibrous. Ox-LDL significantly reduced the viability of human NPCs in a dose and time-dependent manner, and increased the expression of MMP3 induced by LOX-1. Pretreatment with anti-human LOX-1 monoclonal antibody reversed these effects. Ox-LDL, principally mediated by LOX-1, enhanced MMP3 production in NPCs through the NF-κB signaling pathway. In conclusion, increased accumulation of ox-LDL and LOX-1 in IVDs indicates a specific role of the receptor- ligand interaction in degeneration or herniation of IVDs.”

How about a person like myself who's vertebrae degenerated from the radiation and chemotherapy used to defeat cancer? Despite careful application destroying the disease I still ended up "extra crispy" instead of "traditional recipe", finding the bone structure no longer able to support weight.

The VA and Vanderbilt Medical installed an amazing titanium skeletal support structure that's surpassed expectations so much I'm OFF disability and working full time again. Which makes me an idiot - but the work pays better than disability and I can't stand uselessness.

So my laymen question is 'How does this new insight about cellular oxygen usage actually help people?' or is this yet another paper from people trying to publish over perish?

And why DO you post so many medical articles that are obviously geared to those far more specialized in your field than most people here can comprehend? Is it some sort of ego validation thing for your personal benefit or what?

I strive for the opposite, trying to make the complicated understandable to most people in a way that garners their further interest rather than make them feel stupid.

I post write ups that describe something just found that can be useful to people. Your doctors don’t even have this information, unless they also read the breaking news sites, like I do. I only post items people can quickly utilize, as opposed to things yet to come in the future, for the lists I ping.

As I already digested for you, LDL, specifically oxidized LDL, is implicated in back degeneration. As I also stated, reducing LDL or oxidized LDL should be helpful.

The rest is there for support. I already gave you what mattered, so why are you bothered that I posted something immediately useful, from the write up on the study? You don’t have to read a thing but my comments and follow them, to get the “good stuff.”

Is this common surgery now?

Not common at all but they take great effort to monitor me while trying to have me think they're not. I was told that the operation took 14 hours with multiple teams and I may have technically died once during - that persistent aluminum taste and a spasm fit afterward. A nurse asked what was wrong and I said my body just understood the violations of it's sanctity so had a rightful fit of objection.

All good - I'm better than I was and still a fair wage-earner despite eligible for the dole. I don't like seeing the horror on some people when they see the filter band over my throat; though written communication to them on my electronic pad are probably clearer than the verbal ones they're used to.

Am I right or wrong having a 'Handicapped' plate on my car when I can walk better than most? The license people insisted but it doesn't seem right to me.

I don’t see in the article if “concentrated Oxygen” is the same as the Ozone (therapy) I’ve been reading about for IVDD on google scholar.

Is concentrated oxygen the same as ozone ?

The researchers found that higher oxygen levels getting to the back discs actually hurts them, and it appears the back discs encourage more oxygen to come in. Concurrent with this is elevated amounts of oxidized LDL, which might be helping to further change the genes there to get more oxygen, where it's not supposed to be.

What happens is a downward spiral to full degeneration of that damaged disc.

Additionally, our immune system often cannot properly identify oxidized LDL. Sadly, it often sees it as an invader, and the goes after the places that have it—including arteries (atherosclerosis) and back discs (destroying them).

Unfortunately, they seem to talk to an alleviation of pain and some minor heath benefit, but not to the extent of recovering the discs.

Ozone, for what it could affect there, would appear to help oxidate tissues and LDL, which seems counter-productive to the findings in this current study.
