From what I understand if there are any changes in a bill, and there were in this case, it has to be debated and voted on again. So the will need to get the magic 60 one more time. However I could be wrong, the rules of the Senate are weird and arcane.

And there wasn't. The Senate bill went to conference committee.

But now there is a conference report, a "new" entity. Language that has not heretofore been voted on by the senate. And debate on that hasn't been limited by unanimous consent, as far as I know. If there is one objecting senator, to voting tonight, then that objection must be respected. The only way around it is cloture.
