Self pressurization is the next test, using combustion gases from a tap on the one mounted engine, a tap from a turbo pump’s exhaust plumbing fed back to the fuel storage tank, instead of helium or other inert gas.
The thrust puck is outfitted with gimbal mounts for three engines, not just one engine. One Raptor engine produces 440,000 lbs thrust. The Starship empty weight is about 550,000 lbs. No flight is possible with one engine in Earth’s gravity.

Elon Musk himself says the test hop will be with one engine. This isn’t a complete Starship and the fuel will be minimal.
