One thing you mention that does interest me, something about a mechanism for Parkinson’s. When you publish post it on the Board. I would love to read it. Ping me. It would be the first “Medical” literature I have read in years. But the thing I was really interested in during my research was, “What is the mechanism of how the Thalamotomy works?” In the 80s no one knew. Have they figured that out yet?

I asked the same until we saw continued degeneration. Neuroferritinopathy continues in the basal ganglia. The disorder is metabolic.

Why was there an epidemic in the 60’s if not Flu? Sure there are a # of causes of Parkinson’s, drug induced, MPTP or whatever. But there really was an epidemic in the 60s prior to the Pschydelic generation.

I found influenza encephalitis to be the most compelling, however not confirmed. The increase in PD now, including far younger onset is most disturbing.

My hypothesis (model) not including Huntingtons is the neurodegeneration is a metabolic process involving disruption of the sulfur cycle. We have confirmed it with miRNA testing. The primary disruption begins with PANK and involves cysteine, glutathione, cysteamine and hypotaurine. All are related to ROS scavengers. Taurine is secondary to hypotaurine and is causative to sarcopenia wasting and frailty. PKAN is the rosette stone of neurodegeneration.

I think they should have continued the Huntington's trials with cysteamine for longer (increased BDNF in the short trial), however I suspect Huntington's is genetic and not metabolic.

I disturb a few neurosurgeons when I suggest we can eliminate DBS in the near future.

Understand where I'm coming from now?

LOL. Not a bit. That’s probably why my daughter won’t talk neuroscience with me. “Sulfur cycle”? OK, I made “A”s in three semesters of Biochem but Lehninger was a pamphlet in those days.

Didn’t we know Huntington’s involves storage disease due to, can’t remember, funny, I used to know the chemical structure, a complex Hyaluronic something or other? If I recall sulfur was part of it which seemed unusual but maybe not for a glycoprotein.

Ancel Keys. Gave the pseudo intellectual basis for the McGovern Commission on dietary policy.

The sulfur cycle understanding was not really complete until Domini and Stipanuk in 2011. Mostly behind a paywall where it was lost until a few years ago. Cysteamine Dioxgenase filled in the missing pieces to understand PKAN and disruption of the CoA pathway. A forest and trees situation. Everyone looked at it as a pathway. Kind of an intellectual cul-de-sac when it turned out to be a cycle.

40 million euros wasted on research. I'm sure you understand credentialed eminence. They found nothing in the end and drove out the best scientist to Cambridge when it was finished.

He made the most progress and raised the most questions, however maurine models do not recapitulate human brain metabolism.

Hmmm. “McGovern Commission might have been a clue”. I guess that’s why I have never heard of the sulfur cycle.

Our Immunology course was even worse than Lehninger (actually Lehninger was about 400 pages in the first edition, which I still have. Our “White Cell Manual” literally was a Pamphlet. Literally. 60 pages with cardboard cover. LOL. All the early experiments on immunology. It was a lecture during Microbiology. I would imagine Immunology is it’s own course now.

Seven years ago I was new at this. My wife ask me (honey do) to look into PKAN. We found a very large cluster in the Caribbean because of one of her residents.

My professional background was failure analysis and systems studies. It blew up, fell down, shut down or sunk kind of thing. It took me nearly everywhere.

Honey do means do and I liked the change.

I was lucky enough not to have studied the problem conventionally. I went back to the beginnings and that started with Novelli and not Fritz Lipmann. Lipmann was awarded the Nobel prize for the discovery of the CoA pathway. Novelli actually did the work on some of the protein entrances to it. It was this that led us to the cycle. I think Lipmann's discovery was in 1945. People have been stuck in that paradigm. It was very easy for us with miRNA studies to step out of it and show how protein expression was not chiseled in stone and was easily changed.

A very long strange trip.
