There should be a bleeder screw on the side of the valve. Open that with a standard screw driver to release the excess pressure in the tank.
Put your cigarette out before trying this.

Do you happen to have an old regulator - the ones that take a wrench to install around the garage? If so, install it and see if it works. I keep one of those just to use on tank valves that are stubborn.
Is the valve handle moving or not?
If the handle spins call your fire dept and ask about disposal. If its jammed, put it in a chest freezer for an over night. Tap gently when you remove it with a hammer(on the handle straight down) do multiple smacks with increasing force, use a piece of wood in between the handle and the hammer. Try spinning the valve between smacks.
Liquid propane is a liquid, and if water was introduced at the fillup your looking at an ice plug. The colder everything is the brittler they will be.
Make sure if you crack the valve and get a leak you in a place where the tank can be 100 feet plus away from flames and structures. A tarp to throw on top if it leaks might be a good idea

Or do as my buddy and I used to do.... grab the 338 magnum, sit about 300 meters away and shoot it! Great entertainment for the whole family (although not recommended).

I cook with both charcoal and propane grills, depending on what I want to cook.
I have plenty of charcoal and a charcoal chimney to light it and I always have a backup canister of LP.
If you are the cook, make sure your equipment is ready the day before...
I would pick up the tank and drop it about three feet and then try it.
It's worth the $20 to not engulf yourself in flames.

Well assuming it is actually propane and not LNG, I suggest disconnecting it and taking it to Home Depot and exchanging it for another filled tank. Pay the $18 for the exchange and let their propane company worry about it.
Tank exchange is your friend.
If the connection goes INSIDE the bottle valve, then you most likely have a stuck valve in the bottle.
If you have the type that hand screws over the valve on the bottle, you have to have the connection nipple far enough in the bottle valve to release the gas. If the connection fitting doesn't get deep enough into the valve, gas won't come out.
